      <description>Electroporation is a widely used cellular delivery method especially for charged molecules such as DNA, RNA and proteins. Although the term “electroporation” may instill an imagery of pore formation by the electric field, the process is more resembling transient high-energy electrophoresis against the tight cell membrane.The membrane lipid bilayer is almost an insulator with much higher electric resistance compared to the cell plasma. Therefore the electric potential applied on a cell during electroporation is mostly shouldered by the cell membrane and the cell plasma is protected. Macromolecules such as DNA come to a stall after crossing the membrane with the sudden drop of electric field strength. After electroporation, slower cell-autonomous trafficking takes over to distribute the macromolecules to different cellular compartments.Traditionally the electroporation process is known to be highly toxic to the cells from the destruction of cell membranes. However, it is reassuring that any surviving cells would not experience internal damages since cell survival would require membrane integrity and membrane integrity is an insurance for internal protection.Biological membrane composition is not a major factor in the physical process of electroporation. Unlike chemical transfection methods and transduction by viral vectors, electroporation can be performed with all types of cells efficiently and conveniently.Celetrix AdavantageCreatively grouping all mammalian cell types into two groups, each group only needs voltage micro-adjustment. Fast technical support solving Users’ electroporation problems.Limitation of cuvettesWithin the cuvette, the upper portion is a poor area for electroporation as well as the two side portions adjacent to the electrodes.After cutting out the poor areas, the real optimal area (shown as the blue block at the right) in the cuvette is small. For 1mm and 2mm cuvettes, there might be no optimal area at all since the bubbles can spread from the electrodes quite vigorously.Celetrix Electroporation - New Era for T Cell ImmunotherapyPrimary cells transfectionThe Celetrix electroporation technology can electroporate primary cells at very high efficiency. For example, Human PBMC transfection efficiency with GFP plasmid is 64%, and transfection efficiency of CAR-T2A-GFP with SB transposon can reach 60-90%.Human iPSC can be electroporated at a very high efficiency of more than 80% and the cells remain undifferentiated. The Celetrix efficiency is highest compared to all other electroporation methods on the market.Genome editing by CRISPR/Cas9With Celetrix electroporation, we can deliver RNP (ribonucleoprotein complex containing Cas9 protein and gRNA) or Cas9/gRNA plasmid into cells to perform gene editing.The Celetrix™ technology can electroporate human iPSC with Cas9/gRNA. The average efficiency on 4 targeting genes (APP, AAVS1, OCT4 and PDCD1) is more than 50%, and the targeting efficiency on AAVS1 site can be 70%. User friendly system:The eletroporator uses our proprietary electroporation tubes and adapters are used to accommdate different tube sizes.Plug in power cable and switch on the machine. The eletroporator will run a self-test and then proceed to the user interface.Voltage/ Pulse time/ Pulse no./ Pulse interval can be set. We use a single pulse at 30 milliseconds for most cell lines. You'll only need to adjust the voltage. Centrifuge the cells to a pellet and wash the cells once with a serum-free medium.During centrifugation, the electroporation buffer can be prepared. (Celetrix electroporation buffer has 2 components: Part A and Part B. Freshly mix them at 1:1 ratio.)Resuspend the washed cells with the electroporation buffer.Add the plasmid and mix them well.Load the sample and perform electroporation immediately to avoid cell sedimentation.Safety precautions: The metal part underneath the hinged cover and the metal part on the top of the inserted electroporation tube are high-voltage electrical connection terminals. Do not touch them with your hand. While the Celetrix electroporator is straighforward to operate, our electroporation tubes are specifically designed and they must be fully filled with a sample without air bubbles. For example, to load a 120 microliter tube , you'll need to take about 125 microliterl. Insert the pipette tip all the way into the electroporation tube. Rotate the tube and against the pipette tip to wet the bottom. Release the liquid steadily and keep the pipette tip end in the liquid while moving upwards until a bulging convex liquid surface is formed on the top. Press down the tube cover firmly with your thumb till you can feel the metal part on the top slightly sticking out.If there is a tiny bubble after loading, you can still continue as long as the diameter of the bubble is less than 1/3 of the tube. If there is an air bubble larger the 1/3 tube diameter, you'll need to empty the tube and reload it. For the first positive selection, cells are labeled with MACS MultiSort MicroBeads specific for the first marker. After elution from the column, the cells are incubated with MultiSort Release Reagent, which enzymatically removes the MultiSort MicroBeads from the cells. For the second positive selection, the target cells are magnetically labeled with MACS MicroBeads directed against the second marker and isolated on the column.Two consecutive positive selections or positive selection followed by depletionREAlease Technology  allows for magnetic cell isolation by positive selection of target cells and subsequent removal of any beads and labels from the cells. The technology relies on recombinantly engineered antibody fragments instead of whole antibodies for cell surface labeling. Monomeric fragments have a low affinity for cell surface markers. However, when multimerized as part of the REAlease Complex, the fragments bind markers with high avidity. Anti-Biotin MicroBeads bound to the REAlease Complex facilitate isolation of the target cells based on MACS Columns and Separators (see figure). When the column is placed in the magnetic field of the separator, unlabeled cells flow through, whereas the target cells are retained on the column. Subsequently, labeled target cells are eluted from the column using REAlease Bead Release Reagent, which removes Anti-Biotin MicroBeads from the cells.Following elution, there are two options to isolate subsets from this population:I) Disruption of the REAlease Complex by adding REAlease Release Reagent leads to monomerization and spontaneous dissociation of the antibody fragments from the cell surface. Cells are then label free and suitable for magnetic re-labeling with REAlease Technology.II) Alternatively, eluted cells can be directly labeled with MACS MicroBeads.• Bead-free cells: suited for second round of magnetic labeling• Label-free cells: the epitope of a marker becomes completely available again• Recombinantly produced: lot-to-lot consistency allows for reproducible resultsDirect magnetic labelingDirect labeling with MACS MicroBeads is the fastest way of magnetic labeling. MACS MicroBeads specifically bind to antigens on the cell surface. Only one incubation step is required. Direct magnetic labeling requires a minimal number of washing steps and therefore minimizes cell loss. Highly specific cell separation reagents for direct labeling of numerous cell types with MACS MicroBeads are available for human, mouse, rat, and non-human primate cells.Indirect magnetic labelingCombination of primary antibodies and MicroBeadsIndirect magnetic labeling using primary antibodies and MicroBeads is based on a two-step procedure. First, the cells are labeled with a primary antibody directed against a cell surface marker. Conjugated molecules include biotin and fluorochromes. Accordingly, magnetic labeling is achieved with Anti-Immunoglobulin MicroBeads, Anti-Biotin MicroBeads, or Anti-Fluorochrome MicroBeads.This indirect labeling strategy is useful for isolating untouched target cells. In this case, the unwanted cell types are incubated concurrently with a cocktail of primary antibodies conjugated to biotin, for example.
